1) All Wi-Fi contention and data transfer is completely independent of any upper layer protocols. Every transmission must contend for the medium. RTS/CTS isn't actually contention, it is just used to "quiet" the channel.
2) RTS/CTS is configured independently on each STA and AP. ANY Wi-Fi device that receives an RTS will respond with a CTS, no matter how the receiving device is configured.
In your scenario the AP would not initiate an RTS/CTS to the next STA unless the AP was configured for RTS/CTS. So, like I said before, RTS/CTS is configured independently for each device for when they transmit.
